I recently received an item that was missing contents and have been trying to contact the seller. When I messaged the seller there was a popup that said I would have a response within 1-2 days but it's been nearly a week now. So, I was curious, what is the appropriate amount of time to wait before I leave negative feedback and possibly contact ebay/paypal?

You have 45 days to open up a claim with Paypal. Normally, you shouldn't wait any longer than 43 days to open an "Item Not As Described" claim.
You also only have 45 days to leave appropriate feedback and DSR ratings.

I know how long I have to do both, I wanted to know what people on ebay think is an acceptable time to wait for an unresponsive seller to respond before I do through and neg/open a case.

File a not as described dispute through paypal today. Make sure you pay attention to the deadline to escalate to a claim if that becomes necessary. Once you have been given a refund, leave whatever feedback you feel is appropriate. If the seller says they will refund if you close the dispute, tell them you will close the dispute when you get your refund. If they send a transaction cancellation after you are refunded, you can chose to the decline option which prevents them from getting their final value fee credit or you can chose to agree to the cancellation.
You have up to 45 from end of sale to file a dispute and up to 60 days to leave feedbac.
