What's your uptake rate in second chance offers, percentage-wise?
Do you find sending them out immediately after an auction helps?

It's up and down - maybe 50% on average.

I find it to work about 10% of the time and sent immediately after the auction is over. Explanation is up to you, explanation is better with high ticket items not for low price items.

An explanation is always a good idea, because the main use of SCO's is to give the seller an option if th first buyer backs out.
Sellers are not able to determine with certainty that the first buyer has backed out for a minimum of 6 days, so explaining that you have extra pieces tells the buyer WHY you are sending the offer so quick.

More often than not, S.C.O. is used mainly by sellers that have more than one of the same witch i beleive is the case for the OP.
