I listed a fishing reel and got a couple of bids. I went to clean dust off the reel and noticed a fault that would affect the quality. Can I delete the listing and re-list it with the fault detailed?

Cancel the bids and end the listing. Its not fair to ask bidde to put a retraction on their account for an error in your listing..

A note explaining is a good idea.

If you have not notified the Bidder - do so and ask that they cancel the bid, then contact Ebay and ask to cancel the listing due to the issue. DO NOT relist - make it a NEW listing, with the fault highlighted and photos of it.
I have had this happen myself- and most buyers are glad to get the chance to withdraw.
Good Luck!
