Hi,
I am hoping someone could help me out as I am new to ebay selling.
-I sell in Canada on.ca and the US .com and can ship from a Canadian or US Warehouse.
-Is it possible to list 1 item on both and it calculate the shipping depending if it is a CAD buyer or US buyer and ship from the correct warehouse??
Any help would be appreciated.

If you have more than one identical item, you could list
one in US$ on eBay.com offering shipping within the USA only
a second one in Cdn$ on eBay.ca offering shipping to Canada and any other country in the world except the USA
You cannot have identical items available to the same country in the same format (fixed price). That would be a violation of the duplicate listing policy.

If you only have one item: forget it. You can only list it once.
However, considering you are new to selling on eBay, I would keep it simple until you have more eBay selling experience.

I had a similar question last month and know it can be a tricky subject. I asked on Board Hour and this is from the eBay rep

"Keep it simple" is almost ALWAYS the best way to go. It will make your life easier and avoid headaches. Unless you have 2 identical items what you do NOT want is 2 people buying ONE item and also you don't want your account to get into trouble.
