The Standard setting allows you to type (or paste) in plain text. You can then add some fancy stuff (coloured lettering, different fonts) by highlighting and clicking on the options in the tool bar.
The HTML setting allows you to write your description off -site, insert your own coding, and paste it into the Sell Your Item form. You can then click on the Standard tab and see what it looks like "in real life".
