if I decline a "best offer" can I change the price of item upwards ?

if I decline a "best offer" on my item, can I revise the "buy it now"  price of the item  to a higher price as I realised the value is much higher than I previously knew.

You can lower the price but not raise it.

You can end the listing if there are no bids.

If there are bids, you can cancel the bids, and end the listing. In that order! You may be charged fees based on the high bid if you do this.

Remember that if you cancel bids, you will probably never see those bidders again, and that you see only the high bid and not the maximum bid.

If you end the listing you can use Sell Similar to change the asking price or the Best Offer minimum.
