I bought a game that turns out it is a fake and does not play
Can i cancel the order before seller ships? If not, what is my recourse if/when item arrives?

I'm mystified about how you know, before the seller has even shipped, that the game is fake and will not play.
However, you can ask the Seller for a Mutual Cancellation of the transaction.
If you have already paid, make sure you are refunded before agreeing to the MC.
If you have already paid and the item is on its way, on arrival you can explain the situation to the seller and ask for a refund. If he agrees that is the end of it. Destroy the game.
If he tells you to return the game for a refund, use a Delivery Confirmed /tracked service. This will allow you to escalate the dispute, should the seller not refund promptly on arrival of the unwanted item.
Note that tracked services can be expensive. But without them, you cannot enforce the refund.

I am not sure how you know it is a fake and doesnt play if you haven't received it yet. Nonetheless, if the versions listed are not compatible with you DS, you could request a mutual cancellation if the have not shipped it yet.

One other note. If this is the Pokemon game, you are getting free shipping from China. If seller wants it back, assuming it has been shipped, it will cost you double or triple the cost of the game to send it back with tracking.
